چکیده

Exposure to acute psychological or physical stress increases core body temperature (stress-induced hyperthermia, SIH) which is part of the autonomic stress response. SIH is used as a putative rodent anxiety paradigm in which anxiolytic drugs reduce the SIH response. The predictive validity of the SIH paradigm has proven to be good, making it suitable to detect the putative anxiolytic properties of drugs. So far, GABAA receptor agonists including benzodiazepines and hypnotics have proven to attenuate the SIH response. The GABAA receptor has been known to be closely involved in the acute stress response. Also, the recent development of compounds with selective efficacy for different subunits at the benzodiazepine site of the GABAA receptor has renewed interest for the therapeutic potential of GABAergic drugs. Moreover, metabotropic (GABAB) receptor agonists reduce the SIH response. GABAB receptors are ubiquitously expressed in the central nervous system, and there is evidence for a role of the GABAB receptor in anxiety. Thus, both drugs acting on the GABAA and the GABAB receptor are generally able to attenuate the SIH response, and this review presents a detailed overview of the effects of both drug classes on the SIH response. As the GABA receptor family is diverse and complex, this paradigm may contribute to the elucidation of the putative effects of GABAergic drugs in emotional disorders such as anxiety and depression

Fear and anxiety behavior in rats

Fear can be considered as a functional defense behavior to protect living beings against dangerous situation. In our studies we have investigated the fear behavior in rats in elevated plus maze .The increase in two parameters percent of open arm entries (%OAE) and percent of time spent in the open arms (% OAT) and decrease in the percent of time spent in closed arm (%CAT) was considered as fear...

Different effects of tegmental GABAergic receptors on the expression of morphine-induced place preference in rat

In the present study, the effects of excitation and inhibition of GABAA and GABAB receptor subtypes on the expression of morphine-induced conditioned place preference (CPP) were investigated. For this purpose, male Wistar rats (250-300 g) were used in the experiments. Five days after surgical cannulation in the ventral tegmental area (VTA), different doses of morphine were injected into the ani...

POSSIBLE INVOLVEMENT OF GABAA RECEPTOR SITES IN CATALEPSY

The present work was performed to show the possible interaction of GAB A, and GABAB receptors in catalepsy. The following results were obtained: 1- Muscimol (MUS), a GABAA agonist, induced catalepsy. This effect was dose-dependent. Baclofen (BAC), a GABAB agonist, had no effect in this respect. 2- Catalepsy induced by muscimol was potentiated by pretreatment of animals with bicuculline (BI...

P145: The Role of γ-Aminobutyric Acid Receptor in The Social Anxiety Disorder

Social anxiety disorder (SAD) is the one of the most common anxiety disorders. Despite its high prevalence, the disorder is still considerably undiagnosed and untreated. The disease places a massive burden on patient’s lives, affecting not only their social interactions but also their educational and professional activities, thereby constituting a severe disability. γ-aminobutyric acid (GABA) s...
